Synthesis of new long synthetic LowMolecularWeight Heparins - tools to probe heparin biology:Two recent papers in Chemical Science and J. Am. Chem. Soc. describe synthesis of novel long structurally-homogeneous heparanoids, and the utility of these high-value synthetic tools to enable biological investigations. In Chemical Science (2013, 4, 3218-3222; DOI: 10.1039/C3SC51217G), work adapts the Gardiner group’s recent large-scale heparin synthesis to provide higher-O6-sulfated targets through 8-, 10- and 12-saccharide lengths, providing the first examples reported of these pure low molecular weight heparins. These are high-value targets for investigating structural definition of HS-biomolecular interactions, here allowing elucidation structure-specific effects of these saccharides on different growth-factor mediated cellular processes. Additionally, synthetic access to these materials enabled collaboration with Uesugi’s group (Kyoto University), facilitating their investigations of non-GAG synthetic ‘recruiter’ molecules for promoting clustering of HS-bound cell surface syndecan-4, underpinning potential use for cell therapy applications, reported in J.
